Is the Association of single nucleotide polymorphisms of TNF-α-G238A and CCR2-G190A with hypertension in Korean essential hypertensive patients?

Abstract

|
Objective : The G190A polymorphism in CC chemokine receptor 2 (CCR2) and G238A in tumor necrosis factor-α
(TNF-α) gene locus have been shown to be associated with hypertension, coronary heart disease and insulin resistance syndrome, in Caucasians and Japanese. However, because of the difference in the linkage disequilibrium pattern, little information is available on this polymorphism in the susceptibility to hypertension in Korean population. Therefore, the aim of the present study was to evaluate the allelic distribution of TNF-α-G238A and CCR2-G190A genes and to investigate the association of single nucleotide polymorphisms (SNPs) of these two candidate genes with essential hypertension in Korean population.
Methods : Ninety two essential hypertensive patients and seventy three normotensive subjects were genotyped for TNF-α-G238A and CCR2-G190A polymorphisms with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).
Results : There was no association of genotypic distributions between hypertensive and normotensive individuals both at TNF-α-G238A (GG, GA and AA were 95.7%, 4.3% and 0.0% respectively in the hypertensives and 91.8%, 8.2%, and 0.0% respectively in the normotensives; heterozygosity: OR=0.71, 95% CI=0.19 to 2.60, p>0.05) and at CCR2-G190A (GG, GA, and AA were 48.9%, 47.8%, and 3.3% in hypertensives, 57.5%, 38.4%, and 4.1% in normotensives; heterozygosity: OR=1.19, 95% CI=0.62 to 2.23, p>0.05 homozygosity: OR=0.97, 95% CI=0.19 to 5.06, p>0.05) sites.
Conclusions : Our results indicate that TNF-α -G238A and CCR2-G190A polymorphisms might not be associated with essential hypertension in Korean population.
